It is my understanding that this error refers to when someone has typoed a path component to refer to a non-existing directory or a file. As such, this error can frequently occur.

In a message dated 2003-06-23 2:15:07 PM Eastern Daylight Time, bjh21@netbsd.org writes:

# A pathname, with an optional ending slash, that refers to a directory.

But in (e.g.) the error list for open(), we find:

# [EISDIR]
#    A component of the path prefix is not a directory.

Now, by definition, any path prefix must refer to a directory, so this
error can never occur.  This is clearly wrong, so either the definition or
use of the phrase needs to be modified.
